He was a sea captain, operating primarily with two ships, the Marmaduke in 1633 and the Abraham later. He sailed the Atlantic regularly from England to Virginia and also into the Caribbean, primarily to Barbados. He formally emigrated to Virginia in 1635 but continued to sail. In 1638 he acquired a license to sell tobacco in Essex England and became an actual 'factor', rather than just a carrier. He bought land in Virginia in1653 but continued to sail until his death in 1689.

He settled on Chippokes Creek in James City County in 1649. He received 300 acres there for transporting six settlers to Virginia (Cavaliers & Pioneers, Nugent, p.187). About 1652, he appears across the James River in Southwark Parish in Surry County. And later, about 1678, he sells both plantations and moves to his second wife's plantation, Martin's Brandon in Charles City County.

Marriage Notes for JOHN BARKER and ANN: Were married in either 1672 or 1673. Children of JOHN BARKER and LETITIA are:

He must have inherited the land from his father's will which would account for the disposition of the 916 acres of land in his will dated 1714.
