John Hamrick was born in Prince William County by 1744. He was the apparent oldest son of John Hamrick and Sarah (Stone?)

. He married Nancy Hazelrigg in Prince William County by 1770. Nancy was the daughter of William Hazelrigg and Kitty Neavill

. In 1771, John acquired a 134 acre lease in Leeds Manor, Fauquier County, Virginia, near the present-day village of Hume, Virginia, in the Blue Ridge Mountains.

John's and Nancy's children included

John and Nancy were founding members of the Thumb Run Baptist Church in Fauquier County.
