He was most likely born at Hayton Castle, Hayton, Cumberland, England. He served as Deputy Constable of Bewcastle in 1567 (his father was Constable). He lived at Cumcatch, Brampton, Cumberland. His spouse is not known. He was the father of at least two children.

Leonard Musgrave is mentioned in the will of father Thomas Musgrave, dated 20 Feb 1532; in the Inquisition of his father's estate, 12 Oct 1534.; the will of Richard Musgrave, 26 Jan 1552; the will of John Dacre, 3 Mar 1566; and in the inquisition of Leonard Musgrave,10 Jan 1610.
