Any tips on finding mixed-race ancestors?

My paternal grandmother was quite certain that her grandfather (John William Williams) was of Cherokee descent, possibly through his maternal line. It is also possible that he (or his father) was adopted by the Williams family.

Multiracial offspring was very taboo in that area (Ohio) in that time period (Mid-1800s). So, you wonderful researchers, do you have any tips on going about finding the real lineage? Yes, I am aware that a genetic test could give me some ideas about the likelihood of having American Indian ancestry. Sadly, that won't help me much in tracing my family lines.
