Amos married Hannah Allen on March 16, 1780 in Amenia, Dutchess, New York. Together they had the following children: Sarah Allen; Theron S. Allen; Alanson Allen; John S. Allen; Amos Allen, Jr; Abigail Allen; Betsy Allen; Hannah Allen.

He died on February 10, 1822 in Tunbridge, Orange, Vermont, United States and was buried after February 10, 1822 in Tunbridge Meeting House Cemetery, Tunbridge, Orange County, Vermont, USA.
