The son of William Byrd Chamberlayne and Ann Williamson Mosby.

Chamberlayne was a Richmond merchant as well as chief clerk and book-keeper of James River and Kanawha Canal Company.

By 1860 he had established a partnership with Samuel McDearmon in Richmond as McDearmon and Chamberlayne at Shockoe.

Edwin married Sarah Madison Scott on 16 April 1838.

They had ten children, five girls and five boys.
