Mary (Unknown) Gillespie was born circa 1725.

Mary married William Gillespie, Sr. in 1745 in Virginia, United States. Together they had the following children: Mary (Gillespie) Bowen; Simon Gillespie, Sr.; Robert G Gillespie; James "Smiley" Gillespie; John Gillespie; Agnes Gillespie; Rev. Thomas Gillespie; William Gillespie, Jr.; Elizabeth Gillespie.

She died in 1797 in Pendelton, South Carolina, United States.

It has been variously suggested that she was Mary Clendenning, Mary Hughes, or Mary Simpson.

The suggestion she was Mary Clendenning is based on the idea she might have been a sister of Archibald Clendenning, for whom her husband was executor in 1749.

A Mary Gillespie was listed in Archibald Clendenning's will.
