William Holbrook was the son of Erasmus and Betsey (Smith) Holbrook. Educated primarily in public schools he attended Berkshire Medical College in Pittsfield, MA and was an 1848 graduate of the New York University Medical School. Following graduation he commenced practice in Bondsville, MA, then removed to Palmer, MA in 1849.

He married Clara Belknap, daughter of Capt. Peter and Anna, on Feb. 24, 1850. They were the parents of William Edward, born July 25, 1852; Clara B., born Aug. 20, 1856; and Idella Louise, born July 20, 1865.

He was appointed postmaster of Palmer during the administration of President Fillmore in 1852, and in 1858 received the appointment of consulting physician and surgeon of the State Almshouse at Monson.

Dr. Holbrook served on the medical staff of two regiments during the Civil War. He was commissioned and mustered into the 10th Mass. Infantry as an Assistant Surgeon on June 21, 1861. He was subsequently promoted to Surgeon and mustered into the 18th Mass. Infantry on Jan. 13, 1862. He was present with the 18th Mass. Infantry during the 1862 Peninsula Campain, including the Siege of Yorktown, and later that same year at Second Bull Run and Fredericksburg. Holbrook was absent from the Regiment from Sept. 8, 1862 through the beginning of Nov. 1862 due to illness, during which period he was furloughed home. He was absent from March 28, 1863 until he returned in late April 1863, again due to illness. Holbrook was on duty with the Regiment in 1863 at Chancellorsville, Gettysburg, the Mine Run Campaign, and Rappahannock Station. On July 29, 1863 Holbrook was sent to the General Hospital due to a fracture of his left tibula and was absent from the regiment on sick leave from Aug. 3, 1863 to Sept. 22, 1863. In the middle of November 1863 he was promoted to Brigade Surgeon and Chief Operator. He was mustered out of military service on Sept. 2, 1864 at the expiration of his three year enlistment.

Following his military service Dr. Holbrook returned to Palmer, MA and resumed his medical practice which he continued for fifty years. He served on the town school committee for many years, being first elected in 1864, and also served as a member of the board of health. He represented his district in the State Legislature in 1882. He was appointed examiner for State aid, also pension examiner in 1865, and continued until 1892, when he resigned. He held the position as physician for the State Primary School from 1876 to 1886. The appointment of medical examiner, which he held at the time of his death, was conferred by Gov. Rice in 1877, and he was a member of the State Board of Agriculture for six years. He was a member of Thomas Lodge of Masons, and a charter member of L.L. Merrick Post, G.A.R. Post No. 107, serving as the second Post Commander, and attended the 18th Massachusetts Regiment's 40th Anniversary Dinner at Norwell, MA in 1901. He was also a member of state and district medical societies.
