Isaac Lyon was born in 1691 in Newark, Essex County, East Jersey. His parents were Thomas Lyon and Elizabeth Lyon.

Isaac married Hannah Lyon in 1721 in Essex, New Jersey, United States. Together they had the following children:

• John Lyon;
• Mattaniah Lyon;
• Eliphalet Lyon;
• Abigail Woolsey;
• Jane Lyon;
• Unknown Pierson.

He died on February 3, 1764 in Newark, Essex County, New Jersey and was buried after February 3, 1764 in Fairmount Cemetery, Newark, Essex County, New Jersey, United States.
