Joseph was born on Jekyll Island, the son of Henry Charles Poulain du Bignon and Ann Amelia Nicolau.
On January 22, 1839 he married Felicite Elizabeth Riffault, the daughter of Charles Pierre Riffault and Marie Anne Felicite Grand Dutreuilh. They had 6 children:

Josephine, born February 3, 1841 in Jekyll Island, Glynn, Georgia. Married Newton Sobieski Finney.
Henry Riffault, Born June 13, 1843 in Jekyll Island, Glynn, Georgia. Married Alice Madeline Leocadie Symons.
Louise, born abt 1845 in Jekyll Island, Glynn, Georgia
Felicite, born December 4, 1846 in Brunswick, Glynn, Georgia married Judge William Davenport.
John Eugene, born February 2, 1849 in Brunswick, Glynn, Georgia. Married Frances C Schlatter.
In 1839, Joseph was a lieutenant in the Glynn County Volunteers.
In 1845, he was a member of the Georgia House of Representatives.
In 1846, he was a Justice of the Inferior Court of Glynn County.
