There are currently 4 male line descendants of James Rowe SR (via 3 of his 6 sons) who have Y-DNA tested at Family Tree DNA.
1. L. Rowe, Y-37 test, descendant of Stephen Rowe

2. D. Rowe, Y-111 test, descendant of Stephen Rowe

3. S. Rowe, Y-67 test, CTS4466 SNP pack test, descendant of Joseph Duncan Rowe

4. Herbert Roe JR, Big Y 700, descendant of James Rowe JR.

Results from the STR tests (37, 67, 111, and the 700 STRs of the BIg Y) show a relationship consistent with having a recent common ancestor for the 4 men in the right time frame to be James Rowe SR. The CTS4466 SNP pack test result was R1b-A1135 (Z21065). The Big Y 700 SNP result is R1b-A195, consistent with the A1135 result, but a more recent haplogroup mutation. R1b-A195 (at The Big Tree https://www.ytree.net/DisplayTree.php?blockID=2014) is a DNA lineage strongly associated with southern Ireland and the Uí Fidgenti in particular. (FTDNA Uí Fidhgeinte Project https://www.familytreedna.com/groups/ui-fidgente/about/background) This may eventually provide clues to the origin of the James Rowe SR family line in Europe.

The Y-DNA test data, coupled with the genealogical tree and autosomal connections to non-male lines at FTDNA, 23&ME, and Ancestry, such as descendants of the line of Solomon Rowe and multiple descendants of the sister of James Rowe JR, Mary (Rowe) Plymale and her husband James Isaac Plymale, pretty conclusively rules out any NPEs in these lines and proves that James Rowe SR., his progenitors, and all of his descendant males are R1b-A195. Multiple suggestions for the father and brothers of James SR have been suggested on various online genealogical sites over the years, with varying degrees of probability (mostly improbable). In the vacuum of having no secure information for his parentage, misinformation and wishful thinking have filled the void, spreading from site to site like the telephone game. The inclusion of the German George Rowe (Rau) some sites as his father is the prime example of this phenomenon. With this new information it would be rather simple to prove the validity of such suggestions by testing any descendants of these men for comparison.

All Rowe/Roe males who believe they are descended from James Sr through one of his six sons (or even better still, a hypothetical brother, uncle, or cousin so we can finally determine something about James SRs ancestors) are encouraged to join us and take the Big Y 700 Y-DNA test at Family Tree DNA to confirm it. Your participation can also help us further refine the Y-DNA line for James Rowe SR and find his ultimate origins before our family came to the colonies.
