By his first wife Mary Wiggs, he was the father of Samuel, John (died v.p.), Mary (executor of his will in 1655), Elizabeth (Anthony) Olmstead, and Rebecca (Anthony) Montague.

Dr. John Anthony alleged father of John the immigrant in both scenarios, left information carved in stone. His grave marker says: "He dyed ye 28th April 1655, being aged 70 years, and was buried nere this place, and left behind him 1 sone and 3 daughters.[6]
Dr. John Anthony had two sons, Samuel and John and three daughters living in the vicinity of London, named in the 1644 will of Richard Arnold.[7]
John Anthony the immigrant was in Rhode Island at the time. In addition no mention was made of John in the 1655 will of John Anthony, Doctor of Physick.[8]

There is also nothing I see so far to link as siblings these immigrants to America:

The wife Sarah Thornton for Dr Anthony is correct.

The Will of John Anthonie of London Doctor of Physick was dated 21 April 1655 and proved 2 June 1655. He made bequests to his wife Sarah, son Samuel, and daughters Rebecca Montague, Elizabeth Olmstead and Mary Anthonie. He named several friends as overseers and James Smith the younger and Mary his daughter as executors. He left bottles of Aurum potabile to his overseers and to James Smith.[7]

Mary was in England probating his Will in 1655, so couldn’t have been having children in New England.
