Dorothy Cartmell was born in 1660 in England, United Kingdom and was baptized on January 17, 1665 in Painswick, Gloucestershire, England. Her parents were William Pettypool and Ann Pettypool.

Dorothy married Nathaniel Cartmell, Sr. in 1684 in England, United Kingdom. Together they had the following children: Martin Cartmell; Thomas Cartmell; Hannah Cartmell; Nathaniel Cartmell, Jr.; Sarah Cartmell; Nathan Cartmell; Edward Cartmell.

From T K Cartmell's "Early Settlers of the Shenandoah Valley", payge 417: "the widow (Dorothy Cartmell (Poole)) moved to the VA about 1735 with her sons: Martin, Nathaniel, Nathan, and Edward along with several unnamed daughters"

She died on November 1, 1750 in Opeckon, Frederick County, Virginia.
