אלזה שילר נולדה ב-11 בפברואר 1869 באלברפלד (Elberfeld; כיום חלק מן העיר וופרטל) לאביה אהרון שילר, בנקאי פרטי יהודי, ולאמה ז'אנט לבית קיסינג. לאמה נודע אחר כך תפקיד חשוב בשירתה של שילר, בעוד אביה שימש השראה לדמות הראשית במחזה "Die Wupper".
בשנת 1894 נישאה שילר לרופא יונתן ברטולד לסקר (אחיו המבוגר של אלוף העולם בשחמט עמנואל לסקר), ועקרה באותה השנה לברלין. שם פיתחה את כישורי הציור שלה. לאחר כשנה, נטשה לסקר את בעלה ועברה לסגנון חיים בוהמי. מאהבה הידוע הוא יוהנס הולצמן, ממנו נולד בנה פאול[1], ב-24 באוגוסט 1899. בשנה זו החלו להופיע שיריה הראשונים. ספר שיריה המלא הראשון הופיע שלוש שנים אחר כך, בשנת 1902, תחת הכותרת "סטיקס" (Styx).
ב-11 באפריל 1903 התגרשו בני הזוג לסקר-שילר, וב-30 בנובמבר באותה השנה נישאה לסקר-שילר לגאורג לוין, בעל גלריה ומייסד כתב העת 'הסער', לו העניקה לסקר את השם האנגרמי הארווט ולדן. תחת שם זה התפרסם ולדן כאחת הדמויות הבולטות באקספרסיוניזם הגרמני.
בשנת 1906 הופיע ספר הפרוזה הראשון שלה, "ספר פטר הילֵה", הקרוי על שם אחד מחבריה הקרובים ביותר, שנפטר. בשנת 1907 הופיע אוסף הפרוזה "לילותיו של טינו מבגדד". בשנת 1909 פרסמה את המחזה "Die Wupper" (שם הנהר העובר בעיר הולדתה) שלא זכה להצגה על במה בשלב ראשון. עם פרסום אוסף שיריה "הפלאים שלי" משנת 1911 הייתה לסקר-שילר לנציגה בולטת של האקספרסיוניזם הגרמני.
בני הזוג ולדן ולסקר-שילר נפרדו בשנת 1910, והתגרשו רשמית בשנת 1912. ללא פרנסה משלה, נתמכה עתה לסקר-שילר בידי חברים, במיוחד בידי העיתונאי והפובליציסט היהודי-אוסטרי קרל קראוס. עוד בשנת 1912 פגשה לסקר-שילר את המשורר והמסאי גוטפריד בן. נקשרה ביניהם חברות אמיצה והם הפכו לנאהבים[1]. את הרגשתה בנוגע ליחסים אלה ביטאה לסקר-שילר במספר רב של שירי אהבה שהקדישה לו ונענתה בשירי תגובה מצד בן[1]. מותו של בנה בשנת 1927 הביא אותה למשבר עמוק.
בשנת 1932 הוענק לה פרס קלייסט היוקרתי. לאחר עליית הנאצים לשלטון היגרה לסקר-שילר ב-19 באפריל 1933 לציריך, לאחר שהותקפה וסבלה איומים על חייה על ידי נאצים. מציריך נסעה פעמיים לפלשׂתינה-א"י, בשנים 1934 ו-1937. בשנת 1938 נשללה אזרחותה הגרמנית, ובשנה שלאחר מכן נסעה בפעם השלישית לארץ ישראל, כדי להתגורר בירושלים. פריצת מלחמת העולם השנייה מנעה ממנה לשוב לשווייץ.
ימיה בירושלים היו קשים ומלווי סבל ושברון לב. לאחר התקף לב נפטרה לסקר-שילר ב-22 בינואר 1945, ונקברה בבית הקברות בהר הזיתים בירושלים. בתקופת ירושלים המחולקת מצבתה נהרסה. לאחר איחוד ירושלים נאספו עצמותיה לקבר אחים, ועל מקום קברה הונחה מצבה חדשה.
