Wertheimer was born in Bühl, Germany in 1893 and studied in his native town and in Baden Baden. He commenced studying medicine in 1912, initially in Berlin, Bonn and Kiel, before his studies were interrupted by World War I, where he served in a medical capacity in Flanders and Italy, and was awarded the Iron Cross, second class, and other decorations. Following the war, he completed his medical studies in Heidelberg University.[1]
In 1920–21 Wertheimer worked as a doctor at Berlin's municipal orphanage, and subsequently received a position at the Institute for Physiology, University of Halle.
With the Nazi rise to power in Germany Wertheimer lost his job. In 1934 he emigrated to Mandate Palestine and accepted a job as temporary director of the Laboratory of Chemistry, at Hadassah Medical School in the Hebrew University of Jerusalem. He continued working at the Hadassah Medical Center until 1963, and served as dean of the institution in the '50s. Werthimer is regarded as the father in the field of fat metabolism.
In 1956, Wertheimer was awarded the Israel Prize, for medicine.

ורטהיימר נולד בבִּיל (Bühl) שבדרום-מערב גרמניה, ולמד בעיר הולדתו ובבאדן-באדן. בשנת 1912 החל בלימודי רפואה בברלין, בון וקיל. בשנים 1914–1918 שירת בצבא כרופא בפלנדריה ובאיטליה וקיבל על שירותו את צלב הברזל (דרגה שנייה) ועיטור נוסף. בשנים 1919–1920 המשיך ורטהיימר בלימודי הרפואה באוניברסיטת היידלברג וסיים אותם שם
בשנים 1920–1921 עבד כרופא בבית היתומים העירוני של ברלין. לאחר מכן התקבל לעבודה במכון לפיזיולוגיה של אוניברסיטת האלה, ועבד לצד אמיל אבדרהלדן. בשנת 1923 קודם ורטהיימר לדרגת Privatdozent, ובשנת 1927 לדרגת פרופסור חבר. עם עליית הנאצים לשלטון בגרמניה הוא פוטר מעבודתו בעקבות חוק לשיקום שירות המדינה המקצועי, ושקל הצעות עבודה במעבדה לביוכימיה במוסקבה, במחלקה לפיזיולוגיה באוניברסיטת סון-יט-סאן בסין, וכן הצעה לשמש זמנית כמנהל המעבדה לכימיה של בית הספר לרפואה "הדסה" באוניברסיטה העברית בירושלים. על אף האי-ודאות שבמשך התפקיד האחרון, ורטהיימר עלה בשנת 1934 לארץ ישראל וקיבל עליו את התפקיד
עבודתו בבית החולים "הדסה" ובאוניברסיטה העברית נמשכה עד 1963, כפרופסור לפתולוגיה פיזיולוגית ובהיותו ראש מדור הרוקחות (1942–1950) וראש המחלקה לביוכימיה (1953–1963). מחקרו המשותף בשנות הארבעים עם תלמידו בנימין שפירא עסק בתפקיד הליפיד בבני אדם ובצמחים והביא לקביעה בנוגע להיותם של אדיפוציטים כמקור להתפתחות שומן
ורטהיימר זכה בפרס ישראל לרפואה לשנת 1956
