On February 26, 1797, Jacob married Jane Presley in Russell County, Virginia. Jacob and Jane had the following children:

• 1-Catherine Jane Weddington (1802-1869) who married Thomas Jacob Hargis. Catherine died in Sandy Hook, Kentucky but her burial location is unknown.
• 2-Elizabeth Weddington (1804-1843) who married John Lewis Hargis. Elizabeth died and is buried in Breathitt County, Kentucky.
• 3-James Weddington (1806-1874) who married Nancy Fletcher. James died in Pikeville, Kentucky and is buried in Weddington Cemetery (Pike County).
• 4-Jacob Weddington, Jr. (1814-1865) who married Sarah Alice Cassidy. Jacob died in Ashland, Kentucky and is buried in Antioch Cemetery in Christian County, Kentucky.
• 5-Henry Weddington (b. 1816) who married Mary Jane Smith. Henry's date of death and burial location are unknown. There is some evidence that he moved to Illinois.
• 6-John Tazwell Weddington (1816-1851) who married Hannah Meade. John died in Catlettsburg, Kentucky but it is believed he is buried in Pikeville, Kentucky.
• 7-Abigail Weddington (b. 1821) who married Jesse Price Meek. Abbie died some time after 1900 and is buried in Catlettsburg, Kentucky but her date of death is unknown.
• 8-Louisa Weddingtion (b. 1822) who married James William Hillman and Richard Ratliff. Louisa's date of death and burial location are unknown.
• 9-Martha Jane Weddington (1824-1892) who married Reuben Green Meade. Martha died and is buried in Missouri.

On March 6, 1845, Jacob married Parthenia Davis-Hunt, widow of Thomas Hunt, in Pike County, Kentucky. Jacob and Parthenia had the following children:

• 1-William Weddington (1846-1920) who had two children with Nancy Brilla Flanery but married Lettice Horton on March 26, 1865. William and Lettice had 10 children. After Lettie's death, William married Sarah Duvall and they had 6 children. William 'Bill' Weddington is buried in Weddington Cemetery in Sandy Hook, Kentucky.
• 2-Harve Burns Weddington (1849-1914) who married Sarah Elizabeth Banner in 1866 and Mary Jane Vencill-Smith in 1874. Harve died in a duel with Elisha Patrick who also died in the battle. Harve Burns is buried in the Manning Flannery Cemetery in Elliott County, Kentucky.

Parthenia Davis has two children by her first husband, Charles and Francis Hunt. At this time, no further information has been confirmed about them.

On February 16, 1855, Jacob married Catherine Dailey. They had no children.
