Son of Col. James Hamilton and Elizabeth Colepeper. Paternal grandson of the Honourable Sir George Hamilton, 1st Baronet, and Lady Mary Butler. Maternal grandson of John Culpeper, 1st Baron Culpeper of Thoresway, and Judith Culpeper.
James married Elizabeth Reading in 1673. She was the daughter of Sir Robert Reading, 1st and last Baronet, and Jane Hannay, widow of the 1st Earl of Mountrath.
They were parents to 14 children, nine of whom are known to have grown to adulthood and married. One of his sons was lost at sea in 1721, and two others were MPs in the Irish Parliament.
James became Earl of Abercorn on the death of his cousin Charles in Jun 1701. He was created Viscount Strabane and Baron Mountcastle that December.
He was succeeded by his eldest son James. His widow died in 1754.
