Disputed Ancestry
According to to the late Anderson, John Hamilton of Clydesdale, afterwards John Hamilton of Samuelstoun, is the bastard son of James, first Earl of Arran. by a mother not known whose name is. Hamilton Memoirs, p. 84 Stirnet, on the other hand, identifies him as the son of Elizabeth Lindsay. Stirnet: Hamilton 01
Death
John Hamilton of Clydesdale died before 16 February 1558, when his son James Hamilton of Samuelstoun is noticed holding land adjacent to the West Mains of Barfude. Register of the Great Seal of Scotland, A. D. 1546-1580, charter number 2503 on p. 674
Marriage
John Hamilton of Clydesdale married Janet Home, the only child of Alexander, third Lord Home. Hamilton Memoirs, p. 84 With her he got the lands of Samuelstoun, which she possessed in right of her grandmother Dame Nicholas Ker, Lady Home, who was the sole heiress of these lands. Ibidem, pp. 371-72

24 August 1531: A charter by which George, Lord Hume, confirms that he has given the lands of Samuelstoun to his sister Jonet Hume, to be held by Jonet and her lawful heirs male by her nominated husband John Hamilton: "filium naturalem quond. Jacobi com. de Arane dom. Hamyltoun." For greater security Jonet and her husband obtained a charter of confirmation under the great seal of Scotland on 12 February 1531-32 Register of the Great Seal of Scotland, A. 3 February 1550:A charter by which David, bishop of Ross, confirms that he has given the comendatarium of Cambuskenneth to James, Duke of Chatellarault and his son and heir James Hamilton. John Hamilton of Clydesdale is named as one of the heirs of entail: "Johanni H. alias callit Johne of Clyddisdale, fratri dict ducis." Register of the Great Seal of Scotland, A.
