Samuel was baptized on 24 April 1715 at Quincy in the Province of Massachusetts Bay. He was the son of Joseph Nightingale and Hannah Paine.
Joseph died when Samuel was nine, and in his will Joseph made provision for Samuel to be educated through college, if he was so inclined. Samuel graduated from Harvard in 1734. He took his M.A. there in 1737.
He preached for several years, but eventually gave it up for farming in Pomfret, Connecticut, citing poor health as the reason.
He married Abigail Belcher in 1740. Children:

They removed to Providence, Rhode Island in 1751. He built a distillery and became very successful.
He was regularly chosen Justice of the Peace for Providence until 1761, and during the 1760s served intermittently on the Superior Court.
He died at Providence in 1786. The will of Samuel Nightengale Esqr., of Providence, is dated 24 January 1786 and was proved 2 April 1787.[1] It names:
