Prior to Samuel's death, in March 1703/4 he, along with nephew John Case, were appointed co-administrators of the estate of Samuel's brother-in-law John Case of Simsbury. Some two years after Samuel's own death at Hartford, in which he left no extant probate record, by a deed initially dated Feb. 20, 1717/18:

• Know Yee that wee Samuel Spencer of Colchester and Joseph Easton and Sarah his wife & Nathaniel Marsh and Elizabeth his wife, Joseph Cook and Rachell his wife, Cyprian Nickols and Mary his wife, Joseph Symons and Abigail his wife, John Hubbard and Agnes his wife, all of Hartford, do grant, convey, alienate...to John Austin, his heirs, administrators and assigns...a parcel of land in the Town of Hartford... [Hartford Deeds, 3:160.]

The land was obviously the former property of their parents.

No record of burial exists for Samuel, but as his death is recorded in the Hartford First Church records, it is assumed he was interred in the Hartford First Church cemetery, now known as Hartford's Ancient Cemetery, the only cemetery existing in Hartford in 1716 when he died.
