Wenche Klouman debuterte ved Det Nye Teater i 1937 og var senere tilknyttet Den Nationale Scene og i særlig grad Centralteatret. Blant hennes sceneoppgaver var tidstypiske lystspillroller i stykker som Kjære Ruth, Tre må man være og Det kunne vært deg, videre Eliza Doolittle i Pygmalion, operetteroller i Min søster og jeg og Czardasfyrstinnen og en stor, kompleks karakterrolle i den psykologiske thrilleren Mona Lisa-smilet av Aldous Huxley, der hun leverte et oppsiktsvekkende spill. Hun trakk seg tilbake fra scenen tidlig på 1950-tallet.
