William Barnes was a son of James Barnes and Sarah Boone Callaway, the granddaughter of the famous frontiersman Colonel Daniel Boone.

On March 6, 1834 William married his cousin Matricia Callaway in Warren County, Missouri. She was a daughter of Susannah L. Howell and Larkin S. Callaway, the brother of the above Sarah Boone Callaway.
