On January 17, 1955, Eraño Manalo married Cristina Villanueva with whom he has six children. Ten years before his father's death in 1963, Eraño Manalo was elected successor to his father as Executive Minister. As of 2005 his son Eduardo is INC's Deputy Executive Minister.
In 1968 he began his foreign missionary work in Honolulu, Hawaii. His administration is generally deemed good based on growth of the organization under him.
