He was the son of Henry Cobb and his second wife, Sarah Hinckley. He was married in Barnstable on December 20, 1680. His wife was Elizabeth Taylor, daughter of Richard Taylor of Barnstable. Samuel Cobb resided in the lower part of the town, and built a house on the six acre lot that was his father's in the new common field. His first house stood on the southeast corner of the land, on the west side of the lane leading to Indian lands. He soon after built a two story house, a little farther west. It was two stories and constructed in the style common in those days. It was taken down about the year 1805. Samuel & Elizabeth had ten children, all born in Barnstable. Elizabeth died in Barnstable in 1721. Samuel died there on December 27, 1727. Samuel's Gravestone in Cobb's Hill Cemetery, Barnstable, Massachusetts reads: Here Lyes the Body of Mr. Samuel Cob Who Dec'd Sept ye 7th 1727 in ye 73d Year of His Age.
