Judy Dukes has substantial evidence (including DNA) that Samuel Duke was not the son of William but of Robert Dukes of Northampton County:

Samuel Duke was probably born in Isle of Wight County, Virginia about 1722. He likely came to North Carolina with his father.

He first appears in the tax lists in 1743 in both Edgecombe County. He does not appear in land records as frequently as his other relatives.

He was in the Granville Militia in 1754 along with Wm., Wm. Jr., John, Robert and Joseph. He is in the 1755 tax list in Granville County, along with Robert and a William. In 1757 he ordered a survey of 640 acres in Granville County. Also in 1757 he received a grant of 156 acres in Granville County. In August of 1760, he received a 156-acre grant in St. John's Parish, Granville County.

In 1766 he is on the Bute County Tax List with his on Britain (this is indicated on the list). In 1771 he is on the Bute County Tax List alone, and on the Franklin/Warren/Vance Tax List with his sons Britain, Burwell and Sterling. In 1781, he and his sons Starling, Britain and Burwell are Tythables in Warren County. In 1782 and 1783, the same men are Taxable with the addition of Harrell. In 1784, the same men are Taxable with the possible addition of William. On January 1, 1793, Samuel sold 30 acres of land in Warren County to John Blacknell. There is no further record of Samuel.

The name of his wife is unknown.
