Henry was the son of philanthropist Gardner and Mary Low Colby (Roberts), and husband of Mary Lizzie Colby (Chamberlin).
He was educated at Brown University, Providence, Rhode Island (AB 1862, AM 1865, honorary DD 1882); and Newton Theological Institution, Newton, Massachusetts, 1867.
He served as president of the American Baptist Missionary Union (1865–68); held a pastorate at Dayton, Ohio (1868–1903); was president of the Board of Trustees, Denison University; and served as president of the Ohio Baptist Convention (1883–86).
His hymn Waiting on the Eve of Labor was sung at the graduation of his class at Newton Theological Institution, June 26, 1867.

Rev. H.F. Colby, pastor first Baptist Church, Dayton. The gentleman whose name heads this sketch was born in Boston Highlands, Massachusetts, in 1842. He is the son of Gardner Colby a merchant of Boston. Most of his early days were spent in Newton, mass. He graduated from Brown University, Providence, R.I., in 1862. After studying law for some months and spending nearly a year in Europe, he took a full course of study in the Newton Theological Institution, from which he graduated in 1867. In the fall of the same year, he came to Datyon, Ohio, and was ordained to the ministry and became pastor of the First Baptist Church. In this position he has continued fourteen years and has been settled longer in Dayton as a pastor than any other of the ministers in the city. In 1870, he was married to Miss Mary Lizzie Chamberlain, daughter of Edward Chamberlain, Esq., of Boston. He has four children, one daughter and three sons.

Pastor for 35 years of the venerable First Baptist Church of Dayton. He was from a prominent Eastern family and enjoyed a distinguished ministerial career. He ministered to many of Dayton's most influential families (Thresher, Barney, Huffman, McCann, Beaver, Deeds, to name a few) as well as the ordinary people of the town equally. He served on many denominational boards and leadership positions, including what is now Denison University. He is buried in a family lot along with his wife and children and their African-American butler, Julius Caesar Hays.
