Samuel Clayton "jr." (in the earlier Culpeper records until his Uncle Sam Clayton removed to Winchester), was made vestryman, 17 Nov 1766, in place of his father Mjr. Philip Clayton(1702-1786); 24 Nov 1767 he was made church warden of St. Mark's Parish. He served as justice of the peace, sheriff of Culpeper and colonel of Militia when in April of 1775 the Culpeper Minutemen took up arms in defiance of Gov. Dunsmore's seizure of the public powder magazine at Williamsburg.

Along with many others supporting the cause, he was reimbursed for supplies: "17 Jan 1776 Ditto paid French Strother of Colo. Samuel Clayton for Tents furnished the Culpeper Battallion 31.4.6 Ditto paid Ditto for N. Pendleton for Straw furnished said Battalion 2.5.0 Ditto paid Ditto for John Bell for Provisions to Cap'n Green Company 5.19.6"

On the 12th of July 1757 in Essex County, Virginia, Sam married his cousin Ann Coleman, the daughter of Robert (1721-1793) & Sarah (Saunders) Coleman upon whose land Culpeper was founded. They had sons: Mjr. Philip Clayton (1758-1810) who served seven years from the start of the Revolution and married Ann Swann Churchill May 8th, 1799; Sam'l who m. Catherine Price, the daughter of William of Shenandoah County in 1786 ; George Clayton who married Elizabeth Broadus Gaines in 1798; Thomas who married Sarah Cunningham; Robert Coleman Clayton who served in the War of 1812; and a daughter Ann C. "Nancy" who married Jeremiah Strother.
