Princess Sofia is by many historians called the unhappiest of all the children of Gustav Vasa. She was married to Duke Magnus of Sachsen-Lauenburg in a discreet ceremony in the shadow of her brother Eric XIV's wedding to Karin Månsdotter in 1568. The marriage became extremely unhappy; the duke was an alcoholic known to mistreat his subordinates and his servants, and he abused her both mentally and physically in such a violent way that she became insane. It seems that the reasons for this was severe jealousy; according to the cronicle of Aegidius Girs, Magnus: " ...showed his princess all unkindness, spit and shamefull slander, that she of the sorrow was caused great weakness of the head." Princess Sofia lived the rest of her life in seclusion at Ekolsund Castle. She remained mentally unstable, but recuperated enough to run her own household, which she did in a somewhat lively way; she changed head butler twenty-one and housekeeper twenty-three times during these years.

Sofia föddes den 29 oktober 1547 på Stockholms slott som fjärde dotter till Gustav Vasa och Margareta Leijonhufvud. Vid hennes halvbror Erik XIV:s kröning deltog också den artonårige hertig Magnus av Sachsen-Lauenburg, son till hertig Franz, brorson till Gustav Vasas första gemål, Katarina av Sachsen-Lauenburg och alltså kusin till den nykrönte kung Erik. 1566 friade Magnus till den fyra år yngre Sofia. Bröllopet firades den 4 juli 1568, men äktenskapet upplöstes med skilsmässa.

Sofia levde sedan sparsamt och tillbaka draget på Ekolsund tills hon avled nästan utblottad 17 mars 1611. Även Sofia uppvisar drag av sinnessjukdom, som kallades "hufvudsvaghet".

Begravningen skedde i största enkelhet i Stängnäs domkyrka och hon fick varken minnesvård eller epitafium
